Improvement of hyperlipidemia (hypercholesterolemia, dyslipidemia) and treatment of LDL-cholesterol reduction
This drug is used for the treatment of hyperlipidemia, reducing total cholesterol and LDL-cholesterol levels in patients with primary hypercholesterolemia and mixed dyslipidemia, while also lowering Apo-B protein and triglyceride levels and increasing HDL-cholesterol levels. It is also helpful in the treatment of dys-beta-lipoproteinemia and hypertriglyceridemia. It is used particularly in cases of homozygous familial hypercholesterolemia when other treatments are inadequate or cannot be used. This drug is also used as an adjunctive therapy to reduce LDL-cholesterol and Apo-B protein levels in pediatric patients aged 10 to 17 years with heterozygous familial hypercholesterolemia, in conjunction with dietary therapy. It is important to confirm the cause of secondary hyperlipidemia and measure lipid levels before starting treatment. This drug is effective for managing hyperlipidemia and improving cardiovascular disease risk.
For the treatment of hyperlipidemia, it is recommended to initiate treatment with a dose of 20 mg once daily in the evening. Patients with mild to moderate hypercholesterolemia can start with 10 mg. The recommended dosing is 5 to 40 mg taken once daily in the evening, and the dose may be adjusted according to baseline LDL-cholesterol level, treatment goals, and patient response before initiating treatment. Dose adjustments should be made by regularly measuring cholesterol levels at 4-week intervals or thereafter. If cholesterol levels are below the target, reduce the dose. Due to the increased risk of muscle damage including rhabdomyolysis, the use of 80 mg of this drug should be reserved only for patients who have been taking this dose for more than 12 months without evidence of muscle toxicity (refer to Warning under precautions).
The recommended initial dose for pediatric patients (ages 10 to 17) with heterozygous familial hypercholesterolemia is 10 mg once daily in the evening. The recommended dosage range is 10 to 40 mg daily, with a recommended maximum dose of 40 mg per day.
This drug may have side effects affecting the muscles, so if muscle pain or weakness occurs, especially when taking high doses, consult a doctor immediately. This drug may affect liver function, so it should be avoided in patients with liver disease or elevated liver enzyme levels, and periodic liver function tests are important. Women who are pregnant or may become pregnant, as well as breastfeeding women, should not take this drug, and those planning to become pregnant should consult a doctor. When used in combination with certain medications, there may be an increased risk of muscle damage, so patients taking other drugs should inform their healthcare provider. Elderly patients, those with impaired kidney function, and patients with hypothyroidism need to use this drug with caution and should follow their doctor's instructions. This drug contains lactose, so patients with lactose intolerance should not take it. If unexplained muscle pain, tenderness, or weakness occurs while taking this drug, stop taking it immediately and consult a healthcare provider. Some patients may experience side effects such as dyspepsia, headache, constipation, diarrhea, so if symptoms are severe, consult a doctor. Avoid taking grapefruit juice or certain drugs while on this medication, as this may increase the drug's blood concentration and the risk of side effects. This drug can raise blood sugar levels, so patients at risk for diabetes should periodically check their blood sugar. When receiving prescriptions for new medications during treatment, inform your healthcare provider about this drug you are taking. This drug is not for use in children or patients under 10 years of age; pediatric patients should follow a doctor's instructions for use. Store this medication out of reach of children and keep it in its original container.
