Summary

Main use
Treatment of obesity (weight loss and maintenance), reduction and prevention of the risk of weight regain
Who takes it
Adults · Children
Adult dose
The recommended dose of this medicine is 120 mg of orlistat orally 3 times daily, taken with a meal containing fat or within 1 hour after a meal. Patients should continue to follow a nutritionally balanced, mildly hypocaloric diet in which approximately 30 % of calories are derived from fat. Daily intake of fat, carbohydrates, and protein should be distributed evenly among the 3 meals. This medicine may be omitted if a meal is skipped or contains no fat. Because this medicine may reduce the absorption of some fat-soluble vitamins and beta-carotene, patients are advised to take a multivitamin containing fat-soluble vitamins to ensure adequate nutrition. The vitamin supplement should be taken at least 2 hours before this medicine or at least 2 hours after it, such as at bedtime. Taking more than 120 mg of this medicine 3 times daily does not increase its effectiveness. Based on measurements of fat in the stool, the effect of this medicine appears approximately 24 ∼ 48 hours after taking it. After treatment is discontinued, the fat content of the stool generally returns to pretreatment levels within 48 ∼ 72 hours. The efficacy and safety of this medicine have not been evaluated for periods exceeding 4 years. ○Studies have not been conducted in patients with hepatic and/or renal impairment or in elderly patients.
Child dose
Studies have not been conducted in children under 12 years of age.

Precautions

This drug should not be used in patients with chronic malabsorption syndrome or biliary obstruction. Patients with an allergy to this drug or its components should not take it. Pregnant women, women who may become pregnant, and breastfeeding women should not take this medication. If there are signs of liver dysfunction or liver damage, you should immediately inform your doctor and discontinue use of this medication. Patients with hyperoxaluria or calcium oxalate nephrolithiasis should use this drug with caution. This medication can mainly cause adverse reactions in the gastrointestinal system, with symptoms such as abdominal distension, constipation, fatty stools, increased bowel movements, or fecal incontinence. These symptoms usually occur within 3 months of starting treatment and are mostly mild and temporary. However, in some patients, these adverse reactions may persist for more than 6 months. Hepatitis or severe liver damage has been occasionally reported, so if symptoms of liver dysfunction appear, you should inform medical personnel immediately. This drug can reduce the absorption of fat-soluble vitamins (vitamins A, D, E, K) and beta-carotene, so taking a multivitamin supplement is recommended. Vitamin supplements should be taken at least 2 hours before or after this medication to minimize interference with absorption. Taking this medication with a high-fat meal may lead to more frequent gastrointestinal adverse reactions; therefore, it is advisable to distribute fat intake evenly across three meals. When taken with oral anticoagulants, blood coagulation status should be frequently monitored, as this medication may affect the anticoagulant effect. If taken with cyclosporine, the plasma concentration of cyclosporine may decrease, so if using both drugs, the dosing times should be adjusted to at least 3 hours apart. This drug is not recommended for children under 12 years of age, as safety and efficacy have not been established in this population. Pregnant or nursing women should not take this medication, as weight loss during pregnancy may pose risks to the fetus. Serious side effects are rare in case of overdose; however, those who overdose should be monitored for 24 hours.
