This drug helps reduce toxicity caused by folate metabolism antagonists and is used to enhance treatment effect when used with 5-Fluorouracil (5-FU) in patients with metastatic colorectal cancer.

Dosage and Administration

Adult dosage

1. Methotrexate standard therapy: Administer 6-12mg of Leucovorin intramuscularly every 6 hours, four times a day. 2. Methotrexate-Leucovorin rescue therapy: Administer 15mg intravenously every 3 hours for 9 doses and then 8 doses every 6 hours intravenously or intramuscularly. 3. Combined administration with 5-Fluorouracil (5-FU) for metastatic colorectal cancer: Administer calcium levofolinate 200mg/m² per day at minimum.

Precautions

This drug is not suitable for the treatment of pernicious anemia or other megaloblastic anemias due to vitamin B12 deficiency. Even when neurological symptoms persist, caution is required as blood conditions may improve. Glass particles from the injection vial may be introduced, so the vial should be carefully handled when injecting, especially when used in children or the elderly. Women who are pregnant or breastfeeding should take special precautions, and in some cases, administration should be avoided.
