Reduction of toxicity of folate metabolic antagonists and therapeutic effect in progressive colon cancer

  • Toxicity of folate metabolic antagonists
View approved original text

This medication helps to reduce the toxicity that may occur when using folate metabolic antagonists, thereby alleviating the burden on patients. Additionally, it aids in enhancing the therapeutic effect when used with 5-Fluorouracil (5-FU) in the treatment of progressive colon cancer. Such efficacy can be expected from both the injection and tablet forms of this medication. Therefore, it plays a significant role in reducing side effects and improving therapeutic efficacy during cancer treatment.

Precautions

This medication is not suitable for the treatment of pernicious anemia or megaloblastic anemia due to Vitamin B12 deficiency. While neurological symptoms may continue to progress, improvement in blood conditions may also occur, warranting caution. Care must be taken when opening the ampule as glass fragments may enter, particularly when administering to children or the elderly. Pregnant or breastfeeding women should not use this medication in combination with 5-FU, and should use it cautiously if administered alone. Severe allergic reactions such as hives or anaphylaxis may occur due to hypersensitivity; notify medical personnel immediately if unusual symptoms arise. Pain or temporary discomfort may occur at the injection site, and rare instances of fever or central nervous system symptoms may also present. When combined with other anticancer agents, side effects such as diarrhea, skin problems, and stomatitis may arise, necessitating dosage adjustment if symptoms are severe. This medication should be used under the guidance of a specialist, especially during methotrexate treatment, monitoring blood concentration to determine appropriate dosage and treatment duration. Caution is required as drug excretion may be delayed in cases of renal function impairment or fluid retention in the body. There are interactions between this medication and certain anticonvulsants that may increase the risk of seizures, thus dosage should be adjusted while monitoring blood concentrations. Elderly or physically weak patients may have a higher risk of gastrointestinal side effects, requiring special attention. This medication is contraindicated for intrathecal administration, and when administered intravenously, the injection speed should be controlled due to calcium content. Store out of reach of children and keep in the original container to maintain safety and efficacy.
