Adult dosage
This medication is taken once daily, regardless of meals. For patients with primary hypercholesterolemia, the initial dose is recommended at 10/10mg or 10/20mg once daily. For patients who require greater LDL cholesterol reduction (over 55%), the initial dose of 10/40mg once daily is recommended. After starting treatment or adjusting the dose, lipid levels should be checked at intervals of at least two weeks for further dose adjustments.

This medication is used as an adjunct to diet for reducing total cholesterol, LDL cholesterol, Apo B proteins, triglycerides, and non-HDL cholesterol levels while increasing HDL cholesterol in patients with primary hypercholesterolemia and mixed dyslipidemia. It may also be administered in patients with homozygous familial hypercholesterolemia as an adjunct to other lipid-lowering therapies or when other treatments are ineffective. This medication is to be taken in conjunction with a proper diet when diet and non-pharmacological measures alone are insufficient. Prior to treatment, secondary causes such as diabetes and hypothyroidism should be identified and treated as necessary. This medication helps effectively control lipid levels in patients at high risk of cardiovascular events, thus improving the progression of atherosclerotic vascular disease.

Dosage and Administration

Adult dosage

This medication is taken once daily, regardless of meals. For patients with primary hypercholesterolemia, the initial dose is recommended at 10/10mg or 10/20mg once daily. For patients who require greater LDL cholesterol reduction (over 55%), the initial dose of 10/40mg once daily is recommended. Precautions

This medication may cause rhabdomyolysis, a rare side effect that can damage muscles, so patients should immediately notify their doctor if they experience acute muscle pain or weakness symptoms. Patients at risk of impaired kidney function due to severe infections, hypotension, major surgeries or trauma, metabolic disorders, electrolyte imbalances, or uncontrolled seizures should temporarily discontinue or stop this medication. Patients allergic to any components of this medication, those with active liver disease, or those with persistent unexplained elevated liver enzymes should not take this medication. Patients with muscular disorders, pregnant women or women who may become pregnant, and nursing mothers should also refrain from using this medication. Patients taking certain antivirals (glecaprevir, pibrentasvir) and those with lactose intolerance should avoid this medication. Patients with a history of alcohol abuse or liver disease, moderate to severe hepatic impairment, kidney dysfunction or hypothyroidism, those with a family history of muscular disorders, and the elderly, who are at higher risk of muscle damage, should use this medication with caution. Patients taking fibrate medications and those with severe myasthenia gravis or ocular myasthenia gravis should also take extra care. During the course of this medication, side effects such as muscle pain, abdominal pain, and liver function abnormalities may occur, and if these symptoms become severe, patients should consult their physician. Regular liver function tests should be conducted, and treatment should be stopped immediately if symptoms of liver damage (nausea, vomiting, fatigue, etc.) appear. This medication may elevate blood sugar levels; however, its cardiovascular disease prevention benefits outweigh this risk, so blood sugar management should be maintained concurrently. It should not be used during pregnancy or breastfeeding as it may pose risks to the fetus and baby, and women of childbearing age should use appropriate contraception. This medication's safety and efficacy have not been sufficiently established in children and the elderly, so extra caution is necessary. There are many possible drug interactions, so dose adjustment and caution are required when taken with certain antibiotics, antivirals, antifungals, and immunosuppressants. Patients should also be careful as dizziness may occur when driving or operating machinery. In cases of overdose, appropriate treatment should be provided based on symptoms, and liver function and muscle enzyme tests should be performed.
